The development of new DNA detection methods is a requirement in the post-genome era, such as biosafety, human health (hepatitis, AIDS, etc.), which require rapid and convenient DNA or RNA detection technology. Electrochemical technology has the advantages of fast, sensitive, low energy consumption, easy miniaturization and integration, and is considered to be one of the preferred technologies for DNA detection in situations where high-limit requirements such as aging and cost are required. The electrochemical DNA nanobiosensor was developed by Ph.D. student Zhang Wei and others under the guidance of Fan Chunhai, a researcher at the Shanghai Institute of Applied Physics, Chinese Academy of Sciences. Its characteristic is that the nanometer scale of the electrode interface is finely regulated, and the gold nanoparticles are simultaneously introduced for electrochemical signal amplification, thereby significantly improving the sensitivity of DNA detection. The biosensor can quickly detect about 20,000 DNA molecules in 1~2 hours, and the detection sensitivity reaches 10fMol/s DNA, which is about 3 orders of magnitude higher than the conventional fluorescent DNA detection method. The researchers also used a BRCA-1 gene sequence detection model associated with breast cancer to demonstrate that the sensor is highly specific for single-base variability testing (ie, SNP analysis).
